Using antisense oligodeoxynucleotides (ODNs), we investigated whether c-myc protein production by angiotensin II (A-II) was involved in hypertrophy of vascular smooth muscle cells (VSMCs). We designed antisense ONDs specific for c-myc mRNA. The cultured VSMCs were isolated from Sprague-Dawley rats (male 7-8 weeks) by the explant method. The cells were stimulated by 10^<-5> M A-II with or without antisense ODNs or sense ODNs. The growth response to A-II was assessed by the uptake of [^3H]thymidine (Td) or [^3H]leucine. A-II increased [^3H]Td uptake into VSMCs while antisense ODNs for c-myc mRNA specifically inhibited uptake, in a concentration-dependent manner. More than 5pM antisense ODNs showed significant effects on the inhibition and the maximal inhibitory effect was 75% in 500pM concentration. However, sense ODNs did not demonstrate the same results. Similarly, c-myc antisense ODNs decreased [^3H]leucine uptake into VSMCs by A-II stimulation. Our results suggest that the c-myc gene product in VSMCs by A-II is involved in the hypertrophic effects induced by A-II. |
