Relaxation response to calcitonin gene-related peptide (CGRP) on superior mesenteric arteries was markedly enhanced in SHRSP compared to that in WKY. CGRP-induced relaxation was not affected by mechanical removal of the endothelium, or by pretreatment with N^G-nitro-L-arginine (NO synthase inhibitor) in either group. Glibenclamide (K_<ATP> channel blocker) had no influence on CGRP-induced relaxation in either SHRSP or WKY. There were no significant differences between the two groups in the relaxations generated by forskolin, dibutyryl cyclic AMP, and 3-isobutyl-1-methylxanthine. These findings indicate that the increased reactivity to CGRP shown in SHRSP is the results of up-regulation of CGRP receptors in the smooth muscles of the superior mesenteric arteries. |
