The present study investigated whether fibrinolytic activity exists in the liver tissue of both stroke-prone spontaneously hypertensive rats (SHRSP) and Wistar Kyoto rats (WKY). Plasminogen activator (PA) activity in the extract solution of he livers was detected by synthetic substrate (H-D-Val-Leu-Lys-p-nitroanilide dihydrochloride) assay. Total PA activity in the liver extract of WKY (26.8±8.3IU) was approximately 1.5-fold higher than that of SHRSP (18.5±4.3IU, n=8, p<0.005). These PA activities decreased by anti-tissue-type PA (t-PA) IgG and anti-urokinase-type PA (u-PA) IgG. Enzymography of the liver extracts revealed three lytic bands with a molecular weight of 67kDa, 44kDa and 38kDa. The inhibitor activity of the liver extract was detected by the reverse fibrin autography method with one lytic resistance band at 70kDa. Thus, fibrinolytic components were observed in the liver tissue of both strain of rats. |
