内容記述 | Adult T cell leukemia-derived factor (ADF) has been reported to be a cytokine which acts as a defensive factor against oxidative stress. The present study was designed first to assess whether ADF has an antiarrhythmic effect against reperfusion-induced arrhythmias (Experiment 1), second to compare the efficiency of the antiarrhythmic property of ADF with SOD (Experiment 2), and third to examine the effects of the combined administration of ADF and SOD on reperfusion-induced arrhythmias (Experiment 3). The isolated male Wister rat heart (n=364) was subjected to regional ischemia (occlusion of left anterior descending coronary artery for 11 minutes) and reperfusion (3 minutes). Experiment 1 : ADF reduced the duration of ventricular fibrillation (VF) and the incidence of sustained VF, describing a mirrored bell-shaped curve with a dose-dependent response characteristic. The effective dose of ADF (0.25~1.0 mg/l) reduced the duration of VF and incidence of sustained VF from the control values of 160.8±6.8 sec (mean±SEM) and 92.9% to 83.3±20.2 sec~114.7±19.5 sec and 35.7%~57.1%, respectively (P<0.05). Experiment 2 : SOD decreased the duration of VF and the incidence of sustained VF. The dose-response curve exhibited a mirrored bell-shaped profile. The effective dose of SOD (7.5×10^3~1×10^5 U/l) reduced the duration of VF from the control values of 163.5±5.4 sec to 70.8±20.5 sec~113.1±19.9 sec and the incidence of sustained VF from 92.9% to 28.6%~57.1%, respectively (P<0.05). Experiment 3 : The optimal combination doses of ADF and SOD (0.5 mg/l and 5×10^3 U/l, respectively) reduced the duration of VF and the incidence of sustained VF significantly in comparison with those observed with 0.5 mg/l of ADF alone. In conclusion, ADF has an inhibitory effect on the reperfusion-induced arrhythmias and the attenuation of the reperfusion-induced arrhythmias by ADF may arise from the ability of ADF to scavenge hydrogen peroxide. ADF may be clinically useful in the therapy of reperfusion-induced arrhythmias. | |||||||||
