内容記述 | Electroporation is an effective method for inducing poration in the cell membrane and delivering macromolecules into cells in the fields of bioengineering and biomedicine. However, since the internal structures of commercially available power supply devices are not disclosed to the public and exhibit low operability, this study aims to develop an electroporation system utilizing Arduino UNO and an electric relay. This system facilitates the easy generation of waveforms by toggling a pin on and off for specified durations (Arduino programming). In this study, a DC-DC converter was employed as the power supply (DC 45-390 V). The ON/OFF time of the electrical relay was controlled by Arduino UNO for waveform generation. For instance, a waveform with an ON/OFF time of 10/500 ms can be generated. When this waveform was applied to a 3-mm gap electroporation cuvette, an electric field with a field strength of 200 V/cm was produced. Using this setup, 40-kDa fluorescein isothiocyanate (FITC)-dextran was successfully delivered into NIH/3T3 cells, indicating poration in the cell membrane. Thus, this study has established a cell electroporation system with high operability. | |||||||||
