D_3誘導体製剤22-oxa-calcitriol(OCT)のラット7, 12-dimethylbenz(a)-anthracene(DMBA)誘発乳癌に対する抗腫瘍効果,担癌宿主の内分泌動態の変化,および細胞周期に対する影響を実験的に検討した.DMBAにて発癌させたラットを4群に分けOCTを0.1,1.0,または10.0μg/kg週3回,3週間経口群と非投与群(対照群)との間で抗腫瘍効果,内分泌動態,腫瘍の細胞周期について検討を行った.その結果,OCT投与群では対照群に比べ用量依存性に抗腫瘍効果が高くなる傾向がみられた.組織学的所見ではOCT投与群はより均一な腺管の形成とより多くの粘液産生が認められた.Dextran-coated charcoal(DCC)法による腫瘍細胞のestrogen receptor(ER)値は,対照群に比較しOCT投与群で上昇傾向を示した.抗ERモノクロナール抗体を用いた免疫染色ではOCT投与群で一部に染色性の強い部分が散見された.内分泌動態ではOCTは血中estrogenおよびprolactin濃度に影響を与えず,また内分泌臓器(下垂体,副腎,卵巣,子宮)の臓器重量や組織学的所見も対照群と差を認めなかった.さらにOCT投与による血中カルシウム濃度の上昇も見られなかった.Flow cytometryによる細胞周期変化の解析では,OCT投与により腫瘍のS phase fractionは低下し,G_0/G_1 phase fractionは上昇した.以上より,OCTは優れた抗腫瘍効果を有しているが,その作用機序は従来の化学療法剤や内分泌療法剤とは異なり,他剤との併用も含め臨床応用が期待できる薬剤であることが判明した.","subitem_description_language":"ja","subitem_description_type":"Abstract"},{"subitem_description":"本文データはCiNiiから複製したものである。","subitem_description_language":"ja","subitem_description_type":"Other"}]},"item_2_publisher_14":{"attribute_name":"出版者
