{"created":"2023-06-20T16:46:11.679166+00:00","id":17631,"links":{},"metadata":{"_buckets":{"deposit":"85154f14-fa78-4fb8-a915-be354c475622"},"_deposit":{"created_by":29,"id":"17631","owners":[29],"pid":{"revision_id":0,"type":"depid","value":"17631"},"status":"published"},"_oai":{"id":"oai:kindai.repo.nii.ac.jp:00017631","sets":["14:923:1549:4271"]},"author_link":["29753","29755","29754","29752","29751","29756"],"item_2_biblio_info_21":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2016-05-31","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"37","bibliographicPageEnd":"10","bibliographicPageStart":"1","bibliographic_titles":[{"bibliographic_title":"近畿大学生物理工学部紀要"},{"bibliographic_title":"Memoirs of the Faculty of Biology-Oriented Science and Technology of Kindai University","bibliographic_titleLang":"en"}]}]},"item_2_description_33":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"梅ポリフェノール(梅PP)は、梅干製造の過程で大量に生じる梅酢から合成吸着剤を用いて回収したポリフェノール成分を濃縮・乾燥させた粉末である。BALB/cマウスを3群に分け、対照飼料(AIN-93M)、4%梅PP含有飼料、2%ラフィノース含有飼料をそれぞれ自由摂取させ、2週間飼育した。梅PP群では対照群に比較して、盲腸重量および摂食量に有意な増加が見られたが、体重には有意な差はなかった。ラフィノース群でも同様の傾向が見られたが、盲腸重量の増加は有意ではなかった。各マウスの盲腸内容物から全DNAを抽出し、16SリボソームRNA遺伝子のV4領域254塩基対のDNA断片をPCRで増幅させたのち、イルミナMiSeqシステムでシークエンスすることにより腸内細菌フローラを解析した。梅PP群では、硫化水素、アンモニア、インドール、吉草酸などの腐敗臭を生産するとされるClostridium indolis、Oscillospira sp.、Anaerotruncus sp.に代表されるOTU(operational taxonomic unit)の有意な減少が認められた。一方、酢酸や酪酸を生産するとされるAcetatifactor muris に代表されるOTU、難消化性糖を資化して酢酸を生産するとされるRuminococcus bromiiに代表されるOTUが顕著に増加していた。主成分分析の結果、梅PP群はラフィノース群とは傾向の異なるフローラの変化が認められた。これらの結果から、梅PPはラフィノースとは異なる作用メカニズムにより腸内細菌フローラを改善する効果があることが示唆された。\nTo evaluate the effect of ume polyphenol(UPP) prepared from a liquid fraction \"umezu\", the byproduct of Japanese traditional pickled plums \"umeboshi\", on the intestinal microflora, BALB/c mice were fed with AIN-93M diets(control), AIN-93M containing 2%(w/w)UPP, or 4%(w/w) raffinose for 2weeks. The cecal weight of UPP fed mice, but not raffinose fed mice, was significantly heavier than that of the control mice. The V4 region(254bp) of 16S ribosomal RNA gene was amplified from the cecal contents by PCR, sequenced by Illumina MiSeq system, and phylogenetically assigned basically with RDP classifier. UPP group had significantly lower proportion of putrefactive bacterial OTUs (operational taxonomic units), which are represented by Clostridium indolis, Oscillospira sp. and Anaerotruncus sp., than control. In contrast, acetate- and/or butyrate-producing bacterial OTUs represented by Acetatifactor muris and Ruminococcus bromii were remarkably higher compared to the control. Principal component analysis showed that UPP and raffinose had different effects on microflora. We suggest that UPP improves the microflora of intestines by the mechanism different from
