内容記述 | Psoriasis is an important inflammatory disease in the dermatological field. It is characterized with erythematous plaque with thick scales clinically and epidermal thickening and perivascular lymphocyte infiltration histopathologically. Although pathogenesis of psoriasis has not been elucidated fully, abnormalities both in T-cell mediated immunity and keratinization have been addressed. Various cytokines, chemokines, angiogenic factors released from epidermal keratinocytes may affect the innate and acquired immune systems. Interferon α, tumor necrosis factor α(TNF α), interleukin-2 (IL-2), IL-12 and IL-23 have been revealed to be key factors in pathogenesis of psoriasis, a Thl-dominant disease. Then, new biologic drugs targeting these cytokines have been developed recently as T-cell agents, anti-TNF agents and anti-IL-12 and IL-23 agents. This comprehensive review focuses recent findings in pathophysiology of psoriasis and development of new therapeutic modalities with special reference to novel biologics. | |||||||||||||
