vitro におけるPGC の培養が可能である。そのため、PGC はゲノムインプリンティングの消去、精子・卵子形成の過程の研究材料としてよく用いられている。また、PGC をある環境下で培養することで胚性生殖細胞(Embryonic germ cell ; EG 細胞)を樹立することができる(1)(2)。ウサギではin vitro でのPGC の培養系は整っておらず、EG 細胞も樹立されていないため、ウサギでもin vitro でのPGC 培養系の確立、EG 細胞の樹立によって、より正確な発生メカニズムを解明する手がかりになるかもしれない。本研究ではマウスEG 細胞樹立条件を基にウサギにおけるin vitro でのPGC 培養及びEG 細胞の樹立を試みた。まず、マウスEG 細胞樹立と同じ条件でウサギPGC を培養したところ、ウサギPGC 由来コロニーが得られた。そこで、次に、培養系の改良を目的にウサギPGC 培養にはLIF、bFGF、Forskolin の有効性を検討した。その結果、LIF、bFGF、Forskolin を添加することによってアルカリフォスファターゼ(ALP)活性陽性コロニー数が増加し、ウサギPGC 培養においても有効であったことが示唆された。また、得られたコロニーを免疫化学的手法によって評価したところ、マウスEG 細胞と同様にALP 活性、OCT-4、SSEA-1 の発現が確認された。さらにこれらの細胞の分化能を評価するため胚様体、テラトーマ形成の確認を誘起したところ、マウスEG 細胞と同様に胚様体様構造の形成は見られた。しかし、SCID マウスへの移植を行なった結果、テラトーマの形成は確認されなかった。今回得られた細胞は、未分化細胞マーカーであるALP、OCT-4、SSEA-1 を発現し、胚様体様構造の形成能を有していたがテラトーマの形成能は示さなかったことから、PGC からEG
