を強力に排出する性質を示すということを利用した、新たな幹細胞の識別方法が報告されている。この方法により分離される幹細胞は、フローサイトメトリーにおいて SP（ Side-Population）分画として検出される。このような、幹細胞のHoechst33342 排出能力に関与する遺伝子として、ABC（ATP-binding cassette）トランスポーターファミリーのひとつであるBcrp1（Breast cancer resistance protein 1）が報告された。さらに近年、Bcrp1 Exon1 には、3 つのアイソフォーム（A、B およびC）が存在し、造血幹細胞においては分化段階に応じてBcrp1 mRNA アイソフォームの転写に選択性があることが示された。そこで本研究では、まずマウス組織ならびに培養細胞におけるBcrp1mRNA アイソフォームの選択性について検討するとともに、未分化マウスES 細胞におけるBcrp1 mRNAアイソフォームの発現量をReal-time PCR により定量的に解析した。その結果、アイソフォームA が最も高く発現し、アイソフォームC の発現はきわめて低いことが示された。さらに、分化誘導過程におけるBcrp1 mRNA アイソフォームの発現について検討した結果、未分化ES 細胞で高発現しているアイソフォームA およびB は、Oct3/4 やNanog の発現低下に先立ち、分化誘導初期に一度急激に減衰し、その後再び発現が回復することが示された。以上の結果より、Bcrp1 mRNA アイソフォームの発現と選択性はES
