University","bibliographic_titleLang":"en"}]}]},"item_2_description_33":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"哺乳動物の卵巣内には排卵に至らない卵胞が数多く存在する。これらを体外で培養し、発生能力を有する卵子が得られれば、ヒト不妊治療等へ応用できると考えられる。体外発育培養(In vitro growth; IVG)は、卵形成、卵胞形成、卵巣の研究、稀少動物の保護、ヒト不妊治療等を目的として研究されてきた。受精能を有する卵子を得るためには小さく未熟な卵母細胞を発育させ、その後成熟させる必要がある。しかし、発育中の卵母細胞は種特異的なあるサイズに達するまで成熟できず、また種によってその発育期間が異なるためにマウス以外の動物種での研究は不十分であるのが現状である。IVGにおいて、卵子と周囲の細胞との相互作用を維持するために、卵子－顆粒膜細胞複合体 (Oocyte-Granulosa cell Complexes; OGCs)の立体構造の維持が重要となる。2004年、Hiraoらによってウシの体外発育培養培地にPolyvinylpyrrolidone　(PVP)　を添加することで立体構造が維持され、その後の発生率も向上することが報告された。そこで本実験では、ウサギ未成熟卵胞のIVGにおける培養条件の検討をおこない、ウサギ発育途上卵母細胞のIVG後の発生能力に関する検討をおこなった。その結果、ウサギIVGでは8日間の培養が好ましく、培地へのPVP添加がOGCsの立体構造を維持し、2％PVP添加が適している事が示唆された。またその後に、体外成熟培養 (In vitro maturation; IVM)、体外受精 (In vitro fertilization;
