          <datacite:description xml:lang="en" descriptionType="Abstract">Muscle cramps are painful, sudden and involuntary muscle contractions that are generally self-limiting. However, the underlying pathophysiological mechanisms have not been fully elucidated. The objective of this study was to establish a muscle cramp-induced mouse model and to investigate the preventive effect of eicosapentaenoic acid (EPA) on muscle cramps. Male C57BL/6 mice were fed a normal diet with sesame oil, with or without EPA, for six weeks. After four weeks, the muscle cramp group was treated with a vasopressin V₂ receptor antagonist and ammonium chloride. Metabolic acidosis was confirmed by a significant reduction in serum pH and an increase in AQP2 mRNA expression in the kidney of the cramp group. In contrast, EPA administration prevented the decrease in serum pH, increased Akt phosphorylation and shifted MyHC composition toward the slow-twitch muscle fiber type in the soleus muscle. These results suggest that this muscle cramp model may serve as a valuable experimental tool. Furthermore, they indicate that EPA administration may prevent muscle cramps by activating signaling pathways that promote protein synthesis, specifically in slow-twitch muscle fibers.</datacite:description>
