サラシノールをシードとする新規スルホニウム塩型食後過血糖改善剤の合成と活性評価 Synthesis and evaluation of new α-glucosidase inhibitors having thiosugar-sulfonium salt structure based on salacinol 田邉, 元三 タナベ, ゲンゾウ 峯松, 敏江 ミネマツ, トシエ α-グルコシダーゼ阻害剤 サラシノール Salacinol (1) は, アーユルベーダ医学で用いられている糖尿病の特効薬物 (Salacia reticulata) から単離された極めて強い血糖値上昇抑制（α-グルコシダーゼ阻害）作用を示す化合物である. 今回, 1 の５員環チオ糖部が阻害活性に及ぼす影響を検討するために, 1 の５員環を開裂した数種のモデル化合物を合成し, その -グルコシダーゼ阻害活性を評価した. その結果, いずれのモデル化合物の活性も極めて低くとどまり, 強い活性発現には 1 の５員環チオ糖構造が必須であることが判明した. （英文） Salacinol (1) is a potent glycosidase inhibitor isolated from Ayurvedic medicine (Salacia reticulata), which is traditionally used for the treatment of diabetes in Sri Lanka and India. In order to determine the effect of 5 membered thiosugar-sulfonium ring structure of 1 on α-glucosidase inhibition, several seco-analogs of 1 were synthesized.
