The media were MURASHIGE-SKOOG (MS) medium, nitrate (NO3-N) medium, and ammonium salt (NH4-N) medium. We compared the changes in fresh weight, form, bud differentiation, and soluble proteins seen on two-dimensional polyacrylamide gel electrophorersis. The increase in fresh weight was greatest and the changes in form were best on the MS medium, followed by NO3-N medium. Callus growth was inhibited on NH4-N medium. Buds differentiated with both MS and NO3-N media, so an inorganic nitrogen nutrient (ammonium salt) was not essential for such differentiation of poplar callus. Four spots for soluble proteins disappeared on electrophoresis of callus cultured on the NH4-N medium. Their identification might help to explain the effect of inorganic nitrogen nutrients of the ammonium salt type on the metabolism of higher plants.[著者抄録]MS寒天培地を基本とした培地上で、緑色選抜を繰返しつつ、2年に渡り継代培養された一本のポプラ由来カルスを用い、培地中の窒素形態を硝酸態窒素およびアンモニア態窒素のみに変化させた場合のカルスの生重量、形態、不定芽分化率および二次元電気泳動上に現れる可溶性蛋白質パターンの変化を検討した。カルスの生長(生重量、形態)には、硝酸態とアンモニア態の両方の窒素を含むMS培地が最もよく、硝酸態窒素のみの培地がこれに次ぐが、アンモニア態窒素のみの培地では生長が著しく阻害された。オーキシン(2,4-D)濃度を低くすることのみによって、MS培地はもちろん硝酸態窒素のみの培地でも不定芽分化の誘導が可能であり、不定芽分化の誘導にはアンモニア態窒素が必ずしも必要でないことが判明した。二次元電気泳動による可溶性蛋白質の分離により、アンモニア態窒素によって特異的に阻害される4つの多量に存在する蛋白質が明らかとなり、高等植物に対するアンモニア態窒素の影響を探る足掛りがえられた。",
